What is the exact best place to stab someone (from the front) and make sure there will not be enough time to have a second thought about it?

You can only stab once? Go for the throat.

If you can stab multiple times, go for the abdomen and try to pull your knife upwards, perhaps cut a hole in their chest right above the belly button, where the ribcage ends and where you are supposed to perform CPR. Make sure you stab with an upward motion, so that it pierces a lung or heart. Try to reach in the hole if possible and touch and squeeze their organs. That should hurt. A lot.

Cut spleen to sternum. If you can manage the power they will still be standing up when your done. They're organs, blood, all other forms of inards will come out like slicing a hole in the bottom of a sterifoam cup. Instantly.

Exactly what DeadAnztac said. For max damage a spleen to sternum cut going to prevail. This takes a bit more effort than other methods, but is one of the most effective.

The question is... why in the hell do you want to know this?

For the quickest death just go for the skull...
Nobody ever suspects a knife in the skull...
All you need to do is get a good swing, and aim for the side of the skull, above the ear, or aim for the temples...It's fast, and little mess is sustained. You would need a big knife for the spleen - sternum thing, and it's messy...But the death is slow and painless...which is an added bonus...
